Opis ogólny

PDGF (platelet-derived growth factor), is the principal serum mitogen predominantly found in cells of mesenchymal origin. It is expressed in smooth muscle cells, endothelial cells and macrophages.[1] PDGF is localized in α-granules of platelets and released during clot formation. PDGF from human platelets has been purified and described as a cationic glycoprotein (pI 9.5-10.4) having a molecular mass of approx. 30 kDa and composed of two covalently linked subunits, designated as chains A (16 kDa) and B (14 kDa).[2][3] PDGF exists as five different dimers : heterodimers occurs as PDGF-AB and homodimers exists as FDGF-AA, PDGF-BB, PDGF-CC and PDGF-DD).[3][4] In platelets, approx.70% of the PDGF is present as the AB dimer, with most of the remainder as BB. Purified human PDGF shows substantial size heterogeneity, ranging from 27-31 kDa, probably due to the presence of isoforms, glycosylation processing, aging of the platelets, and partial proteolysis during purification.
The PDGF gene is highly conserved and is mapped to human chromosome 7p22.3.[5][6] PDGF is structurally and functionally related to vascular endothelial growth factors.[6]

Zastosowanie

Platelet-Derived Growth Factor-AA human has been used in in vitro transdifferentiation of mesenchymal stem cells into SC (Schwann cells)-like cells.[7]

PDGF (platelet-derived growth factor) not only functions as a growth factor, it also participates in chemotaxis for triggering cell movement. Elevated levels of placental PDGF is observed during late first and third trimester pregnancies. PDGF is associated with wound healing and atherosclerosis.[2] PDGFA mediated PDGFRa (PDGF receptor a) phosphorylation activates pathways associated with cancer. PDGFA has been indicated to induce growth and metastasis in human hepatocellular carcinoma.[8]
The sources of PDGF during wound repair include platelets (predominantly PDGF-AB), smooth muscles (PDGF-A),18 monocyte-derived macrophages (PDGF-B),19 and endothelial cells (PDGF- B).20 PDGF may play a role during normal embryonic development.
Purified PDGF activates two distinct PDGF receptors encoded by separate genes. PDGF-AA binds only to -PDGF receptor but PDGF-AB and PDGF-BB bind to both alpha and β-receptors; i.e., the alpha-receptor binds either A or B chain and the β-receptor binds only the B chain.

HumanKine recombinant human PDGF-AA, expressed in human HEK 293 cells, is a mixture of pro-form, pro-mature, and mature dimers with apparent molecular masses of 35, 40, and 45 kDa, respectively. They are disulfide linked glycosylated homodimers. Production in human HEK 293 cells offers authentic glycosylation. Glycosylation contributes to stability in cell growth media and other applications.
