Biochem/physiol Actions

Methotrexate is an allosteric inhibititor of dihydrofolate reductase (DHFR), the enzyme that catalyzes the conversion of dihydrofolate to tetrahydrofolate. Since tetrahydrolfolate is required for purine and pyrimidine synthesis, methotrexate treatment results in the inhibition of DNA and RNA synthesis.

    1. The solubility of this material in DMSO has not been investigated. Other sources indicate that Methotrexate/Amethopterin is soluble in DMSO at 70 - 90 mg/mL and that stock solutions are stable for 1 month at -20°C and 1 year at -80°C.     1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

    1. Methotrexate is practically insoluble in water.  It does dissolve in solutions of mineral acids (such as HCl) and in dilute solutions of alkali hydroxides. A solution can be prepared in 0.1 M sodium hydroxide at 20 mg/ml (may require some heating).
