General description
Immersion Oil has a defined refractive index of about 1.516. It is important that the refractive index (nD) is about 1.5, similar to that of glass. This enables a homogeneous oil immersion to be achieved. Immersion media are used in conjunction with immersion objectives and are located between the surface of the specimen and the lens of the microscope. Immersion oils practically eliminate light beam deflection so that the effectiveness of the lens is considerably increased.
Application
Immersion oil has been used in the following applications:
- in epifluorescent microscopy and cell identification
- in the assembly and filling of the microinjecting system
- in microscopy to reliably visualize dendritic spines

Immersion oil has a larger impact on 3D resolution. Specifically, matching the immersion oil refractive index to glass, reliably enhances the ability to visualize dendritic spines.

What is the best way to dispose of items with this oil(i.e wipes for objective lenses)? Can they be placed in a bio-hazard waste bin and autoclaved?

Section 13 of the Safety Data Sheet (SDS) indicates that there is "no data available" for waste treatment methods. The GHS classification is Category 4 as a flammable liquid, a Class 2 skin irritant, and Category 1 aquatic hazard. Immersion oil should not be disposed of in the sanitary sewer system. Larger unused quantities should be sent to an authorized waste disposal company. For waste treatment options regarding a few drops of immersion oil on a wipe used to clean a microscope objective, no formal recommendation exists. Local regulations may vary, so contacting the Safety Department for disposal recommendations is advisable.
