The Instructions For Use for NeoClear provides a procedure for use in preparing paraffin embedded sections. Only the paraffin is heated to 60°C. All other steps are performed at room temperature. There is no available information for using NeoClear at 40°C. The Instructions for Use for NeoClear are found on the website's product detail page.
If using for deparaffinization NeoClear is similar in use to Xylene. The user may however need to slightly extend the times for each NeoClear station.
The Instructions for Use state that NeoClear is more sensitive to water. This means it may be necessary to change out the various stations of NeoClear on your staining, compared to changing out Xylene on the staining row.

Is it possible to use NeoClear with paraffin tissue that was initially processed using Xylene?
1 answer-
After embedding the slides in paraffin, Neoclear or Xylene can be used to clear the slides and remove the paraffin before staining. If NeoClear is chosen, NeoMount should serve as the permanent mounting media. It is generally not recommended to use NeoClear and a xylene-based permanent mounting media, as the standard practice in histology is to use a clearing agent that matches the permanent mounting media. If an aqueous mounting media is utilized, the slides are stained, washed in water, and then coverslipped with the aqueous mounting media. In the case of aqueous mounting media, the slides do not undergo NeoClear or Xylene treatment.
