General description

Mammalian homolog of drosophila flightless-1 is encoded by the FLII gene in human (also known as FLI, Fli1, or FLIL). The human flightless-1 homolog (Flii) contains sixteen 23-amino acid leucine-rich repeats (LRR) in its N-terminal half and a gelsolin-like domain (GLD) in its C-terminal half, composed of six segmental repeats typical of many gelsolin family members. Flii is reported to interact with actin and "FLI LRR associated protein" (FLAP) via its GLD and LRR domain, respectively (PMID 9525888). Flii is also shown to interact with the glucose-responsive transcription factor carbohydrate responsive element binding protein (ChREBP) via both the LRR and GLD domains and functions as a negative regulator of ChREBP in cancer cells (PMID 24055811). Increased Flii level is observed in the blistered skin of patients with epidermolysis bullosa acquisita (EBA), and Flii overexpression in transgenic mice results in delayed epidermal barrier formation in embryos and decreased expression of tight junction (TJ) proteins, as well as impaired barrier function among adult transgenic mice (PMID 24375017).
