General description

NADPH:quinone oxidoreductase (NQO1) is an FAD containing electron reductase that regulates the detoxification of quinones obtained from the phenolic metabolites of benzene. It is a homodimeric protein that can bind either of the reducing cofactors; NADPH or NADH . NQO1 regulates chemoprotection, neuroprotection, antitumor quinones and antioxidative defense mechanisms . NQO1 may be overexpressed in breast, colon, pulmonary and hepatic tumors.
NADPH:quinone oxidoreductase (NQO1, DTdiaphorase) is a 2-electron reductase. NQO1 is a FAD-containing protein that exists as a homodimer. It is ubiquitously expressed.

Specificity

Anti-NQO1 (C-terminal) antibody is specific for rat, mouse, and human NQO1. The immunizing peptide specifically inhibits the detection of NQO1 band by immunoblotting.

Immunogen

Synthetic peptide sequence corresponding to amino acid residues 261-274 of human NQO1, with N-terminal added cysteine conjugated to KLH. The corresponding sequence differs by one amino acid in mouse and rat.

Application

Anti-NQO1 (C-terminal) antibody may be used in immunological applications such as immunoprecipitation, immunofluorescence and immunoblotting (approximately 28 kDa).

Biochem/physiol Actions

NADPH:quinone oxidoreductase (NQO1, DTdiaphorase) can bioactivate certain antitumor quinones. NQO1 has a role in antioxidant defense via the generation of antioxidant forms of ubiquinone and vitamin E. High levels of NQO1 gene expression has been observed in liver and lung tumors as compared to normal tissues of the same origin.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
