Gene Information

General description

Mas-related G protein-coupled receptor X2 (MRGPRX2) belonging to the class A GPCR/Mas-related gene family is expressed in sensory neurons, mast cells, and keratinocytes. It has seven transmembrane (TM) α-helices. MRGPRX2 mRNA is found at a high level in the skin. It is also detected in adipose tissue, esophagus, urinary bladder, and lungs. MRGPRX2 gene with two-exon is mapped on human chromosome 11p15.

Immunogen

MRGPRX2 (NP_473371.1, 1 a.a. ~ 330 a.a) full-length human protein.

Biochem/physiol Actions

Mas-related G protein-coupled receptor X2 (MRGPRX2) expressed on mast cells acts as an important receptor for pseudo-allergic responses induced by drugs. It is involved in the evolution of the human nervous system. It is also crucial to mast cells. MRGPRX2 gene mutation impairs the risk of anaphylactic actions facilitated by the MRGPRX2 receptor.

The neuropeptide substance P (SP) mediates neurogenic inflammation and pain and contributes to atopic dermatitis in mice through the activation of mast cells (MCs) via Mas-related G protein-coupled receptor (GPCR)-B2 (MrgprB2, human ortholog MRGPRX2). In addition to G proteins, certain

The human ortholog MRGPRX2 and the mice ortholog, Mrgprb2 are activated by basic secretagogues and neurokinins. A number of commonly used small-molecule drugs (e.g., neuromuscular blocking agents, fluoroquinolones, vancomycin) have been recently shown to activate these receptors under in vitro
