General description

Anti-Angiopoietin-1 detects recombinant human angiopoietin-1. The antibody shows no cross-reactivity with recombinant human angiopoietin-2, recombinant human angiopoietin-4, recombinant mouse angiopoietin-like 3, and recombinant human cornea-derived transcript 6 (CDT-6).The ANG-1 gene is mapped to human chromosome 8q23.1.

Immunogen

purified, NSO-derived, recombinant human ANG-1.

Application

Anti-Angiopoietin-1 antibody may be used for immunoblotting at a working concentration of 1-2 μg/mL. It has also been used for immunoblotting using skeletal muscle, kidney, lung, epididymal fat and brain tissue of mouse. For ELISA, the recommended concentration is 0.5-1.0 μg/mL. The antibody is suitable for immunohistochemistry at a working antibody concentration of 25 μg/mL.Monoclonal Anti-Angiopoietin-1 antibody produced in mouse has been used in Western blotting.

Biochem/physiol Actions

Angiopoietin 1 is an endothelial cell-specific protein that is a ligand for the TIE2 tyrosine receptor. It is required for mature blood vessel formation .
Angiopoietins are growth factors vital for vascular differentiation during normal development. There are four angiopoietins discovered that bind to receptor tyrosine kinases Tie2. Angiopoietin-1 (ANG-1) is a potent pro-angiogenic factor that works by recruits mural cells for endothelial tubule formation and vessel maturation. ANG-1 acts along with VEGF in enhance angiogenesis and increase vascular branching. ANG-1 also affects the inflammatory process and inhibits the capillary transmigration of leukocytes on TNF-α stimulation. Sonic hedgehog, a secreted growth factor, reportedly upregulates the expression of ANG-1. Overexpression of angiopoietins has been observed in a variety of malignancies including neuroblastoma, glioblastoma and lung cancers. While ANG-2 increases the metastasis and angiogenesis of the tumors, ANG-1 inhibits tumor angiogenesis
