General description

Arabidopsis has a complex actin gene family that encodes six protein subclasses. One of the classes is represented by the proteins ACT2 and ACT8, which are similar proteins, but have different flanking sequences, introns and silent nucleotide positions in codons. ACT8 is expressed in root, stems, leaves, flowers, pollen and siliques.
Monoclonal Anti-Actin (plant) antibody recognizes actin in several plant species including Dictyostelium. In Arabidopsis, the antibody recognizes all eight actins isoforms (ACT1, 2, 3, 4, 7, 8, 11 and 12).
Monoclonal Anti-Actin (plant) (mouse IgG2b isotype) is derived from the hybridoma 10-B3 (MabGPa) produced by the fusion of mouse myeloma cells (SP2/0 cells) and splenocytes from BALB/c mice immunized with ACT8 protein. In Arabidopsis, actin is divided into two major phylogenetic classes namely reproductive and vegetative class.

Immunogen

ACT8 protein.

Application

Monoclonal Anti-Actin (plant) antibody produced in mouse has been used in:
  • two-layer phos-tag polyacrylamide gel electrophoresis (PAGE)
  • immunohistochemistry and enzyme-linked immunosorbent assay (ELISA).
  • immunoblotting

Biochem/physiol Actions

In Arabidopsis ACT7 isovariant is essential for normal phytohormone response to hormones in different organs. Mutation in the actin (ACT)1 gene is associated with the development of dwarfism in plants in addition to delayed flowering, highly reduced organ size and varied branching pattern of the leaf trichomes and inflorescence stem. In plants, there is a developmental switch in the modulation of actin isovariants throughout cell differentiation and maturation phase.

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide
