General description

LRRK2 (Leucine-rich repeat serine/threonine-protein kinase 2) is a multifunctional protein belonging to the ROCO family of proteins. LRRK2 contains multiple domains including a ROC (Ras of Complex protein) domain that binds and hydrolyzes GTP, and a kinase domain involved in the phosphorylation of myelin basic protein (MBP), mosein, ezrin, and radixin. LRRK2 itself undergoes autophosphorylation and is targeted by other kinases that modify multiple serine residues at its N-terminus; It is constitutively phosphorylated at Ser935 and maintains this state by interaction with the 14-3-3 protein. Although the kinases upstream of LRRK2 have not been definitively identified, previous studies have suggested that this group of kinases may include PKA. It also contains multiple leucine-rich repeat motifs that facilitate interaction with a range of proteins including PARK2 and PRDX3. Mutations in LRRK2, most of which have enhanced its kinase activity, may contribute to the development of late-onset familial Parkinson’s disease; however, the precise molecular mechanism of LRRK2-induced Parkinson’s disease is still unclear.
