Gene Information

human ... TH(7054)

General description

Tyrosine hydroxylase plays an important role in the physiology of adrenergic neurons. It is the first and rate-limiting enzyme involved in the biosynthesis of the catecholamines Dopamine and Norepinephrine from tyrosine. TH is, therefore, a useful marker for dopaminergic and noradrenergic neurons. The enzymatic activity of TH requires ferrous ions as cofactors and is believed to be regulated by phosphorylation. At least four isoforms of human TH have been identified which result from alternative splicing.

Specificity

Tyrosine Hydroxylase (TH, Tyrosine Monooxygenase). Cross-reacts with all mammalian forms tested to date and some non-mammalian forms.

Immunogen

Native tyrosine hydroxylase from rat pheochromocytoma (J. Biol. Chem.,1982, 257:9416-9423).

Application

Anti-Tyrosine Hydroxylase Antibody is an antibody against Tyrosine Hydroxylase for use in IH & WB.
Immunohistochemistry(peroxidase): A previous lot of this antibody was used at 1:1,000 dilution. The antibody gives specific labeling of noradrenergic axons in primate cerebral cortex (Brain Res., 1989, 500:313-324.).
Note: It is important that you use a secondary antibody that has significant reactivity to sheep IgG1 since the majority of the sheep immunoglobulin is IgG1.
Optimal working dilutions must be determined by the end user.

Quality

Routinely evaluated by Western Blot on mouse brain lysates.

Western Blot Analysis:
1:1000 dilution of this lot detected tyrosine hydroxylase on 10 μg of mouse brain lysates.

Target description

~60 kDa

Physical form

ImmunoAffinity Purified
Purified sheep polyclonal in buffer containing 10 mM HEPES, pH 7.5, 150 mM NaCl, 100 μg/mL BSA and 50% glycerol.

Storage and Stability

Stable for 6 months at -20 to -80°C in undiluted aliquots from date of receipt. Store in a NON-self-defrosting freezer.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variabillity in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.

Analysis Note

Control
Positive: brain (corpus striatum) and adrenal gland. Negative: liver.
