Specificity

Clone 1J14 is a ZooMAb® recombinant rabbit monoclonal antibody that detects CYP11B1. It targets an epitope within 11 amino acids from the N-teminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 11 amino acids from the N-teminal half of human CYP11B1.

Application

Quality Control Testing

Evaluated by Western Blotting in human adrenal tissue lysate.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected CYP11B1 in human adrenal tissue lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound CYP11B1 peptide with at least one hundred thousand-fold (100,000X) higher than with non-specific control peptide.

Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected CYP11B1 in human adrenal gland tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CYP11B1 in human adrenal gland tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Cytochrome P450 11B1, mitochondrial (UniProt: P15538; EC:1.14.15.4; EC:1.14.15.4; also known as CYPXIB1, Cytochrome P-450c11 (Cytochrome P450C11), Steroid 11-beta-hydroxylase, CYP11B1) is encoded by the CYP11B1 (also known as S11BH) gene (Gene ID: 1584) in human. CYP11B1 is a member of the cytochrome P450 superfamily of enzymes, which are characterized by their heme-binding domain crucial for their enzymatic activity. CYP11B1 is a mitochondrial enzyme with a structure that includes a signal sequence that directs it to the mitochondria, where it is integrated into the inner mitochondrial membrane. CYP11B1 is primarily involved in the biosynthesis of corticosteroids, specifically in the conversion of 11-deoxycortisol to cortisol in the zona fasciculata of the adrenal cortex. This enzyme′s activity is essential for the production of cortisol, which plays a key role in glucose metabolism, immune function, and stress response. The expression of CYP11B1 is restricted to the adrenal cortex, reflecting its specialized role in steroidogenesis. Its expression and activity are regulated by adrenocorticotropic hormone (ACTH), which is part of the hypothalamic-pituitary-adrenal axis, responding to physiological stress. CYP11B1 is localized to the mitochondria, specifically within the inner membrane. Overactivity of CYP11B1 or mutations that increase its activity can contribute to conditions such as Cushing′s syndrome, characterized by excessive cortisol production and associated symptoms like hypertension, obesity, and glucose intolerance. Conversely, mutations that reduce CYP11B1 activity can lead to adrenal insufficiency, where cortisol production is inadequate, impacting stress response and metabolism. Additionally, genetic variations in CYP11B1 have been implicated in the modulation of blood pressure and may contribute to the development of hypertension. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
