Shake during hybridization when using the TeloTAGGG Telomerase PCR ELISA

When incubations are performed without shaking, signals may be too low or absent. For best results, follow the guidelines described below.

Telomerase quantification using TeloTAGGG Telomerase PCR ELISA

The TeloTAGGG Telomerase PCR ELISA is used for sensitive qualitative detection of telomerase activity. Absolute readings can vary from experiment to experiment, depending on different factors, including PCR conditions, substrate reaction time, substrate temperature etc. The signals obtained can be compared when a reference sample is used and the results are normalized.

Troubleshooting

The TeloTAGGG Telomerase PCR ELISA is designed for the highly sensitive qualitative detection of telomerase activity. Absolute readings can vary from experiment to experiment, depending on different factors such as PCR, substrate reaction time, substrate temperature etc. Signals obtained in different experiments can only be compared when a reference sample was used in both experiments and results are normalized.

Suitability of the TeloTAGGG Telomerase PCR ELISA for other species than human is dependent on their telomeric repeat sequence. If it is TTAGGG as in humans, this kit should be appropriate. If there is another sequence, there will likely be problems with the primers and it may be necessary to use other primers (for example, the telomerase may not recognize the original kit primers, amplification and hybridization will not be optimal). TTAGGG is the most common telomere repeat found in organisms from the animal and fungal kingdoms. For this kit, Roche is not providing typical results data other than when the human telomeric repeat sequence is present.

When incubations are performed without shaking, signals may be low or absent. Roche therefore highly recommends to following the protocol described above and in the package insert.
