General description

RFC-like DNA damage sensor (RAD17)/checkpoint clamp loader component, a homolog of the Schizosaccharomyces pombe checkpoint gene, rad17 is located on human chromosome 5q13.2. This gene is abundantly expressed in the testis.

Specificity

Anti RAD17 (C-terminal) specifically recognizes RAD17 (70 kDa and 75 kDa).

Immunogen

synthetic peptide corresponding to amino acids 623-640 of human Rad17 (isoform 2), conjugated to KLH via an N-terminal added cysteine residue. The immunizing peptide is present in isoforms 1-4 of RAD17.

Application

Anti-RAD17 (C-terminal) antibody produced in rabbit may be used in:
  • immunoblotting
  • immunoprecipitation
  • immunofluorescence

Biochem/physiol Actions

RFC-like DNA damage sensor (RAD17)/checkpoint clamp loader component participates in DNA replication, repair and modification or chromatin remodeling. Phosphorylation of RAD17 is a critical event during checkpoint signaling in DNA-damaged cells. RAD17 has been reported in colon, breast and non-small cell lung carcinoma. It may also participate in cell cycle control.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
