General description

Ras-related C3 botulinum toxin substrate 1 (UniProt P63000; also known as Cell migration-inducing gene 5 protein, p21-Rac1, Rac-1, Ras-like protein TC25, TC-25) is encoded by the RAC1 (also known as TC25) gene (ORF MIG5; Gene ID 5879) in human. Rac1, RhoA, and Cdc42 (UniProt P63000, P61586, and P60953, respectively) are among the most studied Rho family of small GTPases that are involved in a wide variety of cellular functions, including cell polarity, vesicular trafficking, and cell cycle by regulating intracellular actin dynamics. In mammals, the Rho family consists of 22 members, which can be further divided into the Rac subfamily (Rac1, Rac2, Rac3, and RhoG), Cdc42 subfamily (Cdc42, TC10, TCL, Chip, and Wrch-1), RhoA subfamily (RhoA, RhoB, and RhoC) and other Rho-GTPases (RhoE/Rnd3, RhoH/TTF, Rif, RhoBTB1, RhoBTB2, Miro-1, Miro-2, RhoD, Rnd1, and Rnd2). Akt is reported to negatively regulate mixed-lineage kinase 3 (MLK3) by phosphorylating Rac1 at Ser71.

Specificity

Recognizes Rac1; Strong Rac1 preference with slight Rac2 cross-reactivity.

Immunogen

Recombinant protein containing the full length human Rac.

Application

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Rac1 in NIH/3T3 cells.
The unconjugated antibody (Cat. No. 05-389) is shown to be suitable also for Western blotting, immunohistochemistry, and immunoprecipitation applications.
