Specificity

Clone K28/43 is a ZooMAb® mouse recombinant monoclonal antibody that specifically detects Postsynaptic density protein 95. It targets an epitope within PDZ domains 1 and 2.

Immunogen

Fusion protein corresponding to 223 amino acids from N-terminal half of human PSD-95.

Application

Quality Control Testing

Evaluated by Western Blotting in Mouse brain tissue lysate.

Western Blotting Analysis: A 1:50,000 dilution of this antibody detected PSD95 in Mouse brain tissue lysate.

Tested applications

Western Blotting Analysis: A 1:50,000 dilution from a representative lot detected PSD95 in Rat Brain, Human Brain, Human Hippocampus Membrane, Mouse Brain Membrane, Rat Brain Microsomes, and SH-SY5Y cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PSD95 in rat brain tissue sections.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected PSD95 in rat brain tissue sections.

Flow Cytometry Analysis: 1 μg from a representative lot detected PSD95 in one million SH-SY5Y cells.

Anti-PSD95, clone K28/43 ZooMAb®, Cat. No. ZMS1068, is a recombinant Mouse monoclonal antibody that targets PSD95 and is tested for use in in Flow Cytometry, Immunofluorescence, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Disks large homolog 4 (UniProt: P78352; also known as Postsynaptic density protein 95, PSD-95, Synapse-associated protein 90, SAP-90, SAP90) is encoded by the DLG4 (also known as PSD95) gene (Gene ID: 1742) in human. PSD-95 is a postsynaptic scaffolding protein that plays a critical role in synaptogenesis and synaptic plasticity by providing a platform for the postsynaptic clustering of crucial synaptic proteins. High levels of PSD-95 are observed in postsynaptic density of neurons in the forebrain. It is also present in the presynaptic region of inhibitory synapses formed by cerebellar basket cells on axon hillocks of Purkinje cells. It interacts with the cytoplasmic tail of NMDA receptor subunits and shaker-type potassium channels. It is required for synaptic plasticity associated with NMDA receptor signaling. Overexpression or depletion of DLG4 gene is reported to change the ratio of excitatory to inhibitory synapses in hippocampal neurons. PSD-95 undergoes palmitoylation by DHHC-type palmitoyltransferase 2 (ZDHHC2), which is required for targeting to postsynaptic density, plasma membrane and synapses. Palmitoylation may play a role in glutamate receptor GRIA1 synapse clustering. PSD-95 is reported to undergo rapid synaptic palmitoylation/depalmitoylation cycles during neuronal development. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
