Specificity

Clone 1E12 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Dipeptidyl peptidase 9 (DPP9). It targets an epitope within 21 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the internal region of human Dipeptidyl peptidase 9 (DPP9).

Application

Quality Control Testing

Evaluated by Western Blotting in MCF-7 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected DPP9 in MCF-7 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected DPP9 in lysates from Jurkat and NTERA-2 cells and Human liver tissue..

Affinity Binding Assay: A representative lot of this antibody bound DPP9 peptide with a KD of 6.0 x 10-9 in an affinity binding assay.

Flow Cytometry Analysis: 1 µg from a representative lot detected DPP9 in one million NTERA-2 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected DPP9 in Human liver tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected DPP9 in HT-29 cells.

Target description

Dipeptidyl peptidase 9 (UniProt: Q86TI2; also known as EC:3.4.14.5, DP9, Dipeptidyl peptidase IV-related protein 2, DPRP-2, Dipeptidyl peptidase IX, DPP IX, Dipeptidyl peptidase-like protein 9, DPLP9) is encoded by the DPP9 (also known as DPRP2) gene (Gene ID: 9039) in human. DPP9 is a ubiquitously expressed enzyme that cleaves off N-terminal dipeptides from proteins having a proline or alanine a residue at position 2. Its highest levels are observed in liver, heart, and muscle and low levels are detected in brain. Three isoforms of DPP9 have been described. Isoform 1 is cytosolic and isoform 2 is detected in the nucleus and contains a nuclear localization signal (aa 2-9). A third isoform has also been described that lacks amino acids 832-858. Although DPP9 is associated with mitochondria, the strongest co-localization is observed with microtubules. It participates in a number of cellular processes, including extracellular matrix interactions, cell proliferation, and apoptosis. Knockdown of DPP9 can induce neuropeptide Y-driven tumor cell death, mediated by poly (ADP-ribose) polymerase-1 (PARP-1) and apoptosis-inducing factor (AIF). DPP9 also acts as a key inhibitor of caspase-1-dependent monocyte and macrophage pyroptosis in resting cells by preventing the activation of NLRP1 and CARD8. Silencing of DPP9 is reported to reduce Cbl interaction with Syk. It preferentially cleaves the active form of Syk and acts as a negative regulator to shut-off Syk after B cell stimulation, suggesting that DPP9 can influence how much and how long the B cells respond to stimulation. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
