Quality Control Testing
Evaluated by Western Blotting in A549 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Receptor-type tyrosine-protein phosphatase zeta (PTPRZ1) in A549 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PTPRZ1 in HUVEC and NIH/3T3 cell lysates and a 1:10,000 dilution detected PTPRZ1 in human brain tissue lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PTPRZ1 in human brain tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PTPRZ1 in A549 cells.
Affinity Binding Assay; The binding affinity of this antibody to PTPRZ1-Phosphocan peptide was at least one billion-fold (1,000,000,000X) higher than that obtained with the non-specific control peptide.

Receptor-type tyrosine-protein phosphatase zeta (UniProt: P23471; also known as EC:3.1.3.48, R-PTP-zeta, Protein-tyrosine phosphatase receptor type Z polypeptide 1, Protein-tyrosine phosphatase receptor type Z polypeptide 2, R-PTP-zeta-2) is encoded by the PTPRZ1 (also known as HTPZP2, PTPRZ, PTPRZ2, PTPZ) gene (Gene ID: 5803) in human. PTPRZ1is a single-pass transmembrane glycoprotein that belongs to the receptor-type PTP (RPTP) protein subfamily. Its expression is observed mainly in the central nervous system, where it is localized in the Purkinje cell layer of the cerebellum, the dentate gyrus, and the subependymal layer of the anterior horn of the lateral ventricle. It is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to produce the mature form that contains a large extracellular domain (aa 25-1636), a transmembrane domain (aa 1637-1662) and a cytoplasmic domain (aa 1663-2315). A secreted Isoform is also reported that is generated by shedding of the extracellular domain. PTPRZ1 negatively regulates oligodendrocyte precursor proliferation in the embryonic spinal cord and is required for normal differentiation of the precursor cells into mature, fully myelinating oligodendrocytes. PTPRZ1 is involved in regulating cell adhesion, migration, establishment of contextual memory and axon guidance through the dephosphorylation of tyrosine residues in various signaling pathways. It may also play a role in protecting oligondendrocytes against apoptosis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
