Flow Cytometry Analysis: Staining of one million mouse splenocytes was performed using 5 μL of 1:1 mixture of Cat. No. CWA-1003, Anti-Human CD23 (2G8) ColorWheel® Dye-Ready mAb and CWD-PE, ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of a PE-conjugated Rat IgG2a isotype control.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Low affinity immunoglobulin epsilon Fc receptor (UniProt: P20693; also known as Fc-epsilon-RII, Lymphocyte IgE receptor, CD23) is encoded by the Fcer2 (also known as Fcr2a) gene (Gene ID: 14128) in murine species. CD23 is a homotrimeric, single-pass type II membrane glycoprotein that contains a cytoplasmic domain (aa 1-23); a transmembrane domain (aa 24-49), and an extracellular domain (aa 50-331). The extracellular domain presents striking similarity to C-type lectins. CD23 expression is reported in B cells, monocyte, follicular dendritic cells, platelets, and eosinophils. It is also expressed in cases of chronic lymphocytic leukemia (CLL) and in follicular lymphomas. It serves as a low-affinity receptor for immunoglobulin E (IgE) and CR2/CD21 and plays an essential role in the regulation of IgE production and in B-cell differentiation. In addition to its role as a receptor for IgE, CD23 can also be released from cell surface as a soluble protein (sCD23) via ADAM10-mediated ectoderm shedding. Mice lacking ADAM10 expression in their B cells display defective release of CD23. Clone 2G8 is reported to interact at a site similar to but not identical with the site on CD23 that binds IgE. Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
