General description

Cyclophilin B belongs to the cyclophilin family of immunophilins. It is present in the cytoplasm, nucleus, endoplasmic reticulum (ER)/golgi and plasma membrane. The protein can also be secreted from cells. The associated gene is mapped to human chromosome 15q22.
Monoclonal Anti-Cyclophilin B (mouse IgG1 isotype) is derived from the hybridoma CYPB-3 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide corresponding to the C-terminal region of human Cyclophilin B, conjugated to keyhole limpet hemocyanin (KLH). Cyclophilin B (CypB) is a 21 kDa protein encoded by the peptidylprolyl isomerase B (PPIB) gene that belongs to the cyclophilin family, is a highly conserved class of proteins originally identified as cellular binding proteins for the immunosuppressive drug cyclosporin A. CypB is mainly located within the endoplasmic reticulum (ER) of all cell types.

Immunogen

synthetic peptide corresponding to the C-terminal region of human Cyclophilin B, conjugated to KLH. The corresponding sequence is identical in mouse, monkey and canine and differs by one amino acid in rat Cyclophilin B.

Application

Monoclonal Anti-Cyclophilin B antibody produced in mouse has been used in immunoblotting and immunofluorescence.

Biochem/physiol Actions

Cyclophilin B (CypB) is involved in inflammation, viral infection and cancer. It also plays an important role in protecting cells against endoplasmic reticulum (ER) stress via its PPIase activity. Mutations in the PPIB gene give rise to recessive forms of osteogenesis imperfecta and knockdown of this gene decreases cell growth, proliferation and migration.
In endoplasmic reticulum (ER), Cyclophilin B works as a chaperone for exported proteins. It also participates in the intracellular transport of collagen and proper formation of collagen fibril. Cyclophilin B can bind to the Gag protein of human immunodeficiency type 1 (HIV-1) and helps in HIV replication. It is also involved in the infection of hepatitis C virus (HCV), Japanese encephalitis virus (JEV) and human papillomavirus type 16 (HPV16). Cyclophilin B protects against ER-mediated hypoxia-induced cell death. It is overexpressed in various malignancies.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
