General description

Streptavidin, a 58 kDa protein, is a homotetramer purified from Streptomyces avidinii. It has a high affinity for biotin.[1] Streptavidin has been conjugated with either peroxidase or alkaline phosphatase (ALP). These enzymes are used as protein conjugates because of their comparative stability and a wide range of suitable substrates for different applications. ALP conjugates to streptavidin in immunoassays.[2]

Application

Streptavidin-alkaline phosphatase from Streptomyces avidinii has been used in enzyme-linked immune absorbent spot (ELISpot) to detect interferon (IFN-γ) producing cells.[3][4][5]
The optimal working dilution should be determined empirically using a range of dilutions from a 1 mg/ml stock in buffer.

Unit Definition

One alkaline phosphatase unit will hydrolyze 1.0 μmole of p-nitrophenyl phosphate per min at pH 9.8 at 37 °C.

Lyophilized powder containing trehalose with EPPS, phosphate and traces of magnesium chloride and zinc chloride.
