General description

Tris EDTA buffer is commonly used in molecular biology for reconstitution and storage of many precipitated biomolecules, including DNA and RNA. Tris-HCl is a commonly used buffer and EDTA is a metal chelating agent. TE buffer solubilizes nucleic acids while protecting the molecules from degradation. It is used in nucleic acid isolation, which may be done prior to nucleic acid gel electrophoresis and Northern or Southern blot hybridization.

Application

TE buffer has been used for:
  • isolate, resuspend, or store DNA and RNA for PCR or qPCR[1][2]
  • gel electrophoresis, including Northern and Southern blotting[3]
  • bacterial cells, for AmpC disk test, to measure β-lactamases released from the cells[4]
  • antigen retrieval for immunohistochemistry[3]

Components

TE Buffer contains 1M Tris-HCl (pH approximately 8.0), containing 0.1M EDTA.

Questions

1–8 of 8 Questions
  1. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  3. Can this Tris-EDTA buffer solution be used in an IHC protocol for antigen retrieval?

    1 answer
    1. This product has not been tested specifically for antigen retrieval. The researcher would have to determine suitability. Please see the link below to product E1161. This product is a prepared 10X solution formulated specifically for retrieval and may be better suited the specified application:

  6. What does 'for molecular biology' mean?

    1 answer
    1. The phrase `for molecular biology' means that all lots of this buffer have been tested for DNase and RNase activity, and that no activity was detected.

      Helpful?

  7. What are the final concentrations of Tris and EDTA when Product T9285 is diluted to 1:100 to make it 1X?

    1 answer
    1. When diluted 1:100, the solution is 10mM TRIS with 1mM EDTA.

      Helpful?

  8. What is the shelf life of Product T9285, Tris-EDTA buffer solution?

    1 answer
    1. This buffer solution has a recommended retest date of three years. This information is available on the Specification Sheet, and on the lot-specific Certificates of Analysis.
