General description

C24:1 Ceramide (d18:1/24:1) is a synthetic ceramide derived from sphingosine with nervonic acid (a monounsaturated analog of lignoceric acid (24:0)). C24:1 Ceramide-d7 (d18:1-d7/24:1(15Z)) is a deuterated derivative of C24:1 Ceramide (d18:1/24:1).

Application

C24:1 Ceramide-d7 (d18:1-d7/24:1(15Z)) has been used as a standard in liquid chromatography–mass spectrometry (LC-MS) for quantitative analysis of C24:1 ceramide in skeletal muscle. It has also been used as a standard to measure C40-C44 ceramides in zebrafish brain and human embryonic kidney cell line (HEK293) by thermo q-exactive plus quadrupole-orbitrap mass spectrometery.

Biochem/physiol Actions

Monitoring the levels of the ceramides (Cer) d18:1/16:0, Cer d18:1/18:0, Cer d18:1/24:0, and Cer d18:1/24:1 and ratios in human plasma aids the prognosis of fatal outcome of coronary artery disease. C24:1 Ceramide (d18:1/24:1) in a stable isotope form is suitable for a newly described validated liquid chromatography-tandem mass spectrometry methodology for their clinical-scaled measurement.

Insulin sensitivity (as determined by a hyperinsulinaemic-euglyceamic clamp) decreased 15% after reduced activity. Despite not fully returning to baseline physical activity levels, insulin sensitivity unexpectedly, rebounded above that recorded before 2 weeks of reduced physical activity by 14% after the recovery
