General description

The gene encoding protein transport protein Sec23 homolog B (SEC23B) is about 54kb long and located on the chromosome 20p11.23. It is composed of five domains: zinc finger domain, trunk domain, β-sheet domain, helical domain and gelsolin domain.

Application

Biochem/physiol Actions

Protein transport protein Sec23 homolog B (SEC23B) is part of the coat protein complex II (COPII) which is required by proteins to be transported out of the endoplasmic reticulum (ER). It is a secretion-associated RAS-related protein 1 (SAR1) GTPase-activating protein and also plays a role in cargo recognition. Conversion of SAR1-GTP to SAR1-GDP causes its dissociation from the COPII coat. SAR1-bound SEC23B interacts with the C terminus of SEC31, creating a link between the outer-coat SEC13–SEC31 complex and the inner-coat SAR1–SEC23B–SEC24 complex. When it interacts with the outer coat, the GAP activity is stimulated and this accelerates the GTP hydrolysis of SAR1. SAR1 gets released and SEC23B interacts with the trappin protein transglutaminase (TRAPPI) complex subunits like dynactin which are all involved in vesicle tethering, fusion and transportation along microtubules. Finally, SEC23 is phosphorylated and this leads to the fusion of COPII vesicles with the target membrane.
