General description

B-cell lymphoma/leukemia 11B (UniProt Q99PV8; also known as B-cell CLL/lymphoma 11B, BCL-11B, COUP-TF-interacting protein 2, mRit1, Radiation-induced tumor suppressor gene 1 protein) is encoded by the Bcl11b (also known as Ctip2, Rit1) gene (Gene ID 58208) in murine species. CTIP2/CBL11B is a C2H2 zinc finger protein that binds GC-rich response elements and is reported to associate with a variety of cofactors, including the corepressor complexes NuRD, Sirt1, histone acetyltransferase (HAT) p300, functioning both as a transcriptional repressor and activator. CTIP2/CBL11B is required at multiple developmental checkpoints for T cell commitment and maintenance. CTIP2/CBL11B expression is initiated at double-negative 2 (DN2) stage of T cell development and is maintained in mature T lymphocytes, with lower levels found in NK cells, but not in B cells, myeloid or dendritic cells. Bcl11b-/- mice die shortly after birth due to a blockade of thymocyte development at the DN2 stage. Bcl11b-deficient DN2 thymocytes are highly proliferative and retain the potential to differentiate into NK cells and myeloid lineages, but not mature T-cells.

Specificity

Clone 25B6 recognizes both the Alpha and Beta spliced isoforms of human and mouse CTIP2/BCL11B.

Immunogen

GST-tagged recombinant full-length mouse CTIP2 Beta isoform (Senawong, T., et al. (2003). J. Biol. Chem. 278(44):43041-43050).

Application

Anti-CTIP2/BCL11B Antibody, clone 25B6, Cat. No. MABE1045, is a highly specific rat monoclonal antibody that targets CTIP2 and has been tested in Immunofluorescence, Immunohistochemistry, and Western Blotting.
Immunohistochemistry Analysis: A 1:240 dilution from a representative lot detected CTIP2/BCL11B in mouse brain tissue sections.

Immunofluorescence Analysis: A representative lot detected CTIP2 localization in 4% paraformaldehyde-fixed floating embryonic (E14 & E16) and postnatal (P3 & P6) mouse brain sections (Arlotta, T., et al. (2005). Neuron. 45(2):207-221).

Western Blotting Analysis: A representative lot detected doublet CTIP2 bands, representing two spliced isoforms, in Jurkat nuclear extracts as well as in SIRT1 immunoprecipitate (Senawong, T., et al. (2003). J. Biol. Chem. 278(44):43041-43050).

Quality

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected CTIP2/BCL11B in 10 µg of HEK293 cell lysate.

Target description

~96 kDa observed. 95.52/94.57 kDa (human/mouse isoform Alpha) and 88.48/87.34 kDa (human/mouse isoform Beta) calculated. Uncharacterized bands may be observed in some lysate(s).
