The cell line A375-C6 has been shown to be sensitive to the IL-1 mediated anti-proliferative effect. It has been derived from the parental line A375 (Sigma Catalogue number. 88113005) by limiting dilution together with its twin clone A375-C5 (Sigma Catalogue number. 97011320) which is IL-1 unresponsive. A375-C6 could be useful for a growth inhibition assay for IL-1. The cells have been eradicated from mycoplasma at ECACC.
Split sub-confluent cultures (70-80%) 1:5 i.e. seeding at 1-3 x 10,000 cells/cm2using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
