Application

GAP-27 has been used as a blocking peptide for Cx43 (connexin 43) gap junctions in FDC (follicular dendritic cells)-B cell cultures.

Biochem/physiol Actions

GAP-27 shares homology with a region of extracellular loops of connexins, and hence, is thought to inhibit junctions formed by the connexins Cx32, Cx40 and Cx43. It highly hinders co-operative cell-cell interactions including the synergized beating of embryonic cardiomyocytes. This peptide reduces osteoclastic activity and survival rates. This peptide significantly reduces acetylcholine-mediated hyperpolarization of smooth muscle cells.
Inhibitor of gap junction signaling in endothelial cells.

Follicular dendritic cells (FDC) show homo- and heterocellular metabolic coupling through connexin 43 (Cx43) gap junctions and support B cell selection and maturation in germinal centers. In follicular lymphomas B cells escape apoptosis while FDC develop abnormally. Here we tested

The nature of the vasodilator endothelium-derived hyperpolarizing factor (EDHF) is controversial, putatively involving diffusible factors and/or electrotonic spread of hyperpolarization generated in the endothelium via myoendothelial gap junctions (MEGJs). In this study, we investigated the relationship between the existence of

Bone remodelling is dependent on the balance between bone resorbing osteoclasts and bone forming osteoblasts. We have shown previously that osteoclasts contain gap-junctional protein connexin-43 and that a commonly used gap-junctional inhibitor, heptanol, can inhibit osteoclastic bone resorption. Since heptanol
