蔗糖是一种二糖,蔗糖酶水解葡萄糖和果糖。葡萄糖是细胞呼吸中用于能量产生的主要碳水化合物。当细胞摄取时,葡萄糖能够被糖酵解分解或聚合用于能量储存。

應用

葡萄糖和蔗糖检测试剂盒已被用于:
  • 测定蔗糖、有氧糖酵解和丙酮酸激酶活性
  • 测定草莓中的蛋白质和糖
  • 测定小鼠血浆胰岛素和葡萄糖水平
  • 分析血浆葡萄糖和血浆乳酸并定量测定葡萄糖/蔗糖

適合性

原則

在该检测试剂盒中,葡萄糖被葡萄糖氧化酶氧化,生成的比色(570nm)/荧光 (λex = 535 nm/λem = 587 nm) 产物和葡萄糖含量成正比。为了测量蔗糖,会在反应中加入蔗糖酶,将蔗糖转化成葡萄糖和果糖。总葡萄糖减去自由葡萄糖就是蔗糖的浓度。该试剂盒检测线性范围,葡萄糖荧光分析是0.2-1.0nmole,葡萄糖比色分析是2-10nmole。
