General description

Streptavidin, a biotin binding protein produced by the bacteria Streptomyces avidinii, is a very high affinity and specificity non-covalent biotin binding protein that is used in a wide range of biochemical/immunological assays such as western blot and ELISAs. It is a component of the streptavidin-biotin system; wherein biotin is conjugated to a target molecule that is later detected by a signal tagged streptavidin. Streptavidin may be covalently tagged with a range of enzymes that develop chromogenic sustrates or fluorescent molecules to support detection systems.
β-galactosidase may be used in genetic, molecular and immunologic detection systems. Active β-galatosidase cleaves the detection molecule X-gal (5-bromo-4-chloro-indolyl-β-D-galactopyranoside) to form an intense blue product that may be detected by chromogenic techniques.

Specificity

Streptavidin–β-Galactosidase conjugate is reactive with biotin labeled molecules.

Application

Streptavidin–β-Galactosidase conjugate may be used in conjunction with biotin labeled molecules in techniques such as ELISA, immunohistochemistry, protein blotting or with biotinylated DNA probes. It provides a convenient yet highly sensitive and specific detection system.
Streptavidin–ß-Galactosidase conjugate has been used in the fusion assay in chinese hamster ovary cells.

Physical form

Contains Tris buffer salts.

Preparation Note

Streptavidin and β-galactosidase grade VIII are conjugated by a modification of the method of O′Sullivan and purified by gel filtration.
