Specificity

Clone 1D10 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects E3 ubiquitin-protein ligase RNF213. It targets an epitope within 24 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal half of human E3 ubiquitin-protein ligase RNF213.

Application

Quality Control Testing

Evaluated by Western Blotting in A549 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected RNF213 in A549 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected RNF213 in lysates from Human umbilical vein endothelial cells (HUVEC) and A431 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected RNF213 in A549 cells.

Affinity Binding Assay: A representative lot of this antibody bound RNF213 peptide with a KD of 3.3 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected RNF213 in Human cerebral cortex and Human colon tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

E3 ubiquitin-protein ligase RNF213 (UniProt: Q63HN8; also known as EC:2.3.2.27, ALK lymphoma oligomerization partner on chromosome 17, Mysterin, RING finger protein 213, RING-type E3 ubiquitin transferase RNF213) is encoded by the RNF213 (also known as ALO17, C17orf27, KIAA1554, KIAA1618, MYSTR) gene (Gene ID: 57674) in human. RNF213 is a widely expressed, homo-oligomeric cytosolic protein that is reported to catalyze ubiquitination of both proteins and lipids, and is involved in various processes, such as lipid metabolism, angiogenesis, and cell-autonomous immunity. It acts as a key immune sensor by catalyzing ubiquitination of the lipid A moiety of bacterial lipopolysaccharide (LPS) via its RZ-type zinc-finger and restricts the proliferation of cytosolic bacteria. It is also involved in the non-canonical Wnt signaling pathway in vascular development. It acts by mediating ubiquitination and degradation of FLNA and NFATC2 downstream of RSPO3, which leads to the inhibition of the non-ca

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
