Application

In addition to protein determination and quantification in solution,[1][2][3] the QuantiPro BCA Assay Kit has other applications, including determination of protein covalently bound to agarose supports and protein adsorbed to multiwell plates. It has also been applied to the determination of functional groups such as sulfhydryl, N-hydroxysuccinimido carboxylate, aldehyde and hydrazide on a variety of solid supports.
The kit can be used to measure very dilute protein concentrations in very small sample volumes. Accurately measures protein concentrations from 0.5 to 30 μg/mL in tube assays and 1 to 20 μg/mL in 96- or 384-well plate assays.

Principle

Based on the alkaline reduction of Cu2+ to Cu1+ by proteins, and the formation of a bicinchoninic acid:Cu1+ complex having an absorbance maximum at 562 nm.

Storage and Stability

Store Reagents QA, QB, and QC at room temperature. Reagents QA and QB can be mixed together without Reagent QC (4% Copper(II) Sulfate Pentahydrate Solution 12 mL) and stored in a closed container at room temperature for a week. If this kit is received or stored cold, a precipitate may form in Reagent QA or Reagent QB. To dissolve the precipitate, warm the solution slowly to room temperature while mixing. The solutions are now suitable for use.

  1. How many samples (1 mL) can be analysed with the kit?

    1. Below is the link to the product bulletin. The reagents and their volumes included in the kit can be found on page 1. Table 1 on page 2 shows the volume of QuantiPro Working Reagent to prepare and the amount of each component needed:

      For 15 2 ml Standard Test tube assays, 7.75 ml of QA, 7.75 ml of QB, and 0.31 of Copper(II) Sulfate Reagent QC are needed. By calculation, about 480 assays can be done. Fewer may be gotten due to loss during pipetting. Also one may use more or less Protein Standard Solution depending upon how many assays they do each time. Protein Standard Solution (P0914) is available separately in case one needs more for their assays.

  2. How many samples can be analyzed for one kit ? How long can the reagents be kept and at which temperature ?

    1. This kit can be used in several different formats. The format chosen, the number of blanks, and standards will determine the number of tests. The protein standard requires refrigerated storage. Other components are stable up to 1 week at room temperature.
