Biochem/physiol Actions

Remodelin is an inhibitor of acetyl-transferase NAT10, a nucleolar N-acetyltransferase involved in stabilization of microtubules. Remodelin was found to correct cell defects associated with progeria, improving nuclear shape and reducing the DNA damage believed to be associated with mutations in the gene for laminin A. Although the mechanism is still being studied, microtubule stability appears to be involved. Remodelin inhibition of NAT10 activity in laminopathic cells appears to result in reduced microtubule anchorage, normalizing the nuclear shape of laminopathic cells. Remodelin should be a useful tool to study how NAT10 affects nuclear architecture and its relationship to and possible treatments for laminopathies and aging and as well as its over-expression found in a variety of soft tissue sarcomas.
