Especificidad

Clone 2L21 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects HIF-2 / EPAS1. It targets an epitope within 22 amino acids from the C-terminal half.

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal half of human HIF-2 /EPAS1.

Aplicación

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HIF-2 /EPAS1 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected His/ABP-tagged recombinant fragment of Human HIF-2 /EPAS1.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HIF-2 /EPAS1 in HepG2 cells.

Affinity Binding Assay: A representative lot of this antibody bound HIF-2 /EPAS1 peptide with a KD of 1.1 x 10-8 in an affinity binding assay.

Enzyme Immunoassay (ELISA) Analysis: Serial of dilutions from a representative lot detected His/ABP-tagged recombinant fragment of Human HIF-2 /EPAS1.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Endothelial PAS domain-containing protein 1 (UniProt: Q99814; also known as EPAS-1, Basic-helix-loop-helix-PAS protein MOP2, Class E basic helix-loop-helix protein 73, bHLHe73, HIF-1-alpha-like factor, HLF, Hypoxia-inducible factor 2-alpha, HIF-2-alpha, HIF2-alpha, Member of PAS protein 2, PAS domain-containing protein 2) is encoded by the EPAS1 (also known as BHLHE73, HIF2A, MOP2, PASD2) gene (Gene ID: 2034) in human. HIF-2 is a transcription factor that is involved in the induction of oxygen regulated genes. It displays about 48% identity to HIF-1 and is widely expressed with high expression observed in endothelial cells, placenta, lung, and heart. Its levels are strongly induced by hypoxic conditions. HIF-2 dimerizes with ARNT, via amino acid sequence 171-192, and this heterodimer binds to core DNA sequence 5′-TACGTG-3′ within the hypoxia response element (HRE) of target gene promoters. Forma física

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitución

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Almacenamiento y estabilidad

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
