Application

Sodium palmitate (PA) has been used:
  • to induce inflammation and thrombosis pathway in murine macrophage cell line RAW 264.7 cell line by activating reactive oxygen species (ROS) production, Janus-kinase (JNK) signalling and release of histone H3 by western blotting and cell viability by MTT assay
  • to induce lipogenesis in AML12 cells and primary hepatocytes to analyse the effect of irisin on PA induced lipogenesis and related signal pathways by western blot analysis and quantitative PCR analysis
  • as a component in free fatty acid mixture to induce cellular steatosis in HepG2 cell lines and determination of lipid accumulation by Oil-Red-O staining

Biochem/physiol Actions

Sodium palmitate is the sodium salt of palmitic acid, a component in hard soaps. Palmitic acid is a common saturated fatty acid and produced during fatty acid synthesis. Sodium palmitate enhances lipogenesis, cellular steatosis in various cell lines. Palmitate induces cell death in human epidermal growth factor receptor 2 (HER2)/neu-positive cells and breast cancer cell lines like MCF-7 due to enhanced fatty acid accumulation. Sodium palmitate induces lipoapoptosis in L02 and HepG2 liver cells by inducing glycogen synthase kinase-3β (GSK3β) expression.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   3. How long is the dissolved solution stable for ie dissolved in methanol at ~10-50 mg/mL? Do you recommend storing at -20C once dissolved?

    1. Unfortunately, the solution stability of this material is not determined. In such situations, it is recommended to use freshly prepared solutions. However, stock solutions of product P5585, the free acid form, are stable at -20°C when prepared at 20 mg/mL in DMSO or ethanol and purged with inert gas (argon or nitrogen). This salt form may demonstrate similar stability.

  4. Is there any other solvent such as DMSO or ethanol in which this compund is soluble ? For use in cell culture is there any specific recommendation so as to palmitate enter the cell, such as conjugation to fatty acid free BSA ? Thank you in advance

    1. There are no specific solubility tests for this item performed in DMSO or ethanol. The quality control is performed at 50 mg/mL in MeOH, and no specific usage for this application is recommended. It would be necessary to refer to specific literature on preparation for use in cell culture. The following article does provide some guidance for the preparation of this item in sterile water dissolved at high heat in a serum free DMEM media with BSA. Please refer to the article citation below and consider a further literature search if this does not meet your intended requirements.

    1. Sodium palmitate does not dissolve readily in methanol at room temperature.  We test the solubility by heating it for up to 15 minutes to get a clear, colorless solution. Note that the boiling point of methanol is approximately 65°C.
