Specificity

Clone 1G21 is a ZooMAb® rabbit recombinant monoclonal antibody that detects BCL2/adenovirus E1B 19 kDa protein-interacting protein 3 (BNIP3). It targets an epitope within 18 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the N-terminal half of human of human BNIP3.

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected BNIP3 in Jurkat cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected BNIP3 in Human kidney tissue lysate and 1:10,000 dilution detected His/ABP-tagged recombinant fragment of Human BNIP3.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected BNIP3 in Human kidney tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound BNIP3 with a KD of 7.9 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected BNIP3 in U2OS cells.

Enzyme Immunoassay (ELISA) Analysis: A serial dilutions from a representative lot detected His/ABP-tagged recombinant fragment of Human BNIP3.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

BCL2/adenovirus E1B 19 kDa protein-interacting protein 3 (UniProt: Q12983; also known as BNIP3) is encoded by the BNIP3 gene (Gene ID: 664) in human. BNIP3 is an atypical member of the BCL-2 family and the pro-apoptotic BH3-only protein subfamily. Its N-terminal region contains a PEST domain, a BH3 domain (aa 100-125) and a conserved domain. The C-terminal region includes four major domains and involved in protein-protein interactions. The transmembrane domain (aa 164-184) allows for dimerization and insertion into membranes to elicit cell death. The function of BNIP3 ranges from differentiation, growth, maturation, proliferation, stress responses, activates mitochondrial autophagy and initiating numerous forms of cell death through various pathways. It may play context- and cell-type specific roles. BNIP3 expression is widespread in various tissues, including the heart, skeletal muscle, liver, and brain. Its expression is often upregulated under conditions of cellular stress, such as hypoxia (low oxygen levels), ischemia-reperfusion injury, or nutrient deprivation. Dysregulation of BNIP3 expression has been observed in a range of cancers. BNIP3 has also been associated with cardiac diseases, including heart failure and myocardial infarction. Additionally, BNIP3 has been implicated in neurodegenerative diseases, such as Parkinson′s disease. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
