Biochem/physiol Actions

Vacquinol-1 (NSC13316) is a brain penetrant inducer of catastrophic vacuolization in glioblastoma cells (GCs). Vacquinol-1 displays high cytotoxicity to GCs; it induces cell death by membrane ruffling, cell rounding, massive macropinocytic vacuole accumulation, ATP depletion, and cytoplasmic membrane rupture of GCs. Vacquinol-1 originally was developed as an antimalarial compound.
Vacquinol-1 dihydrochloride is potentially used as a therapeutic for Glioblastoma multiforme (GBM). This drug is highly stable in vivo, with a half-life in plasma of 52 hour. Vacquinol-1 has high oral bioavailability and good brain penetration.

In situ transgenesis methods such as viruses and electroporation can rapidly create somatic transgenic mice but lack control over copy number, zygosity, and locus specificity. Here we establish mosaic analysis by dual recombinase-mediated cassette exchange (MADR), which permits stable labeling
