Specificity

Clone 1B20 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Protein disulfide-isomerase A4 (PDIA4). It targets an epitope within 22 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal half of human Protein disulfide-isomerase A4 (PDIA4).

Application

Quality Control Testing

Evaluated by Western Blotting in A549 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PDIA4 in A549 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PDIA4 in NIH3T3, Panc-1, and PC12 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PDIA4 in NIH3T3 cells.

Affinity Binding Assay: A representative lot of this antibody bound PDIA4 peptide with a KD of 3.3 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PDIA4 in human liver tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Protein disulfide-isomerase A4 (UniProt: P13667; also known as EC:5.3.4.1, Endoplasmic reticulum resident protein 70, ER protein 70, ERp70, Endoplasmic reticulum resident protein 72, ER protein 72, ERp-72, ERp72) is encoded by the PDIA4 (also known as ERP70, ERP72) gene (Gene ID: 9601) in human. Protein disulfide isomerases (PDIs) are a family of multifunctional enzymes with oxidoreductase and chaperone activities that regulate the redox state of proteins and maintain their proper folding and function. The PDI family has 21 members and PDIA4 is the biggest of the PDIs with three CGHC motifs, two in the N-terminal and one in the C-terminal half. It is a part of a large chaperone multiprotein complex. It is synthesized with a signal peptide (aa 1-20), which is subsequently cleaved off to produce the mature protein that mainly resides in the lumen of endoplasmic reticulum (ER) where the KEEL sequence at its C-terminus prevents its secretion from ER. PDIA4 is implicated in the growth and death of tumor cells and its expression is shown to be upregulated in a variety of tumor cell lines and in human lung adenocarcinoma tissues. Its levels are also upregulated during ER stress. It is reported to promote lung cancer development via regulation of STAT3/VEGF axis in T and B lymphocytes of cancer stroma. It is reported to regulate the activation and degradation of procaspases 3 and 7 by caspase 8. It controls the stability and function of these caspases in a CGHC motif-dependent manner. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
