Target description

Nonstructural protein NS8 (UniProt: A0A6B9VTP3; also known as ORF8 protein) is encoded by the orf8 (also known as NS8) gene (Gene ID: 43740577) in Sars-CoV-2. Coronaviruses (CoV) represent a diverse family of positive-sense RNA viruses capable of causing respiratory and enteric disease in human and animal hosts. Amongst these the SARS-CoV-2 is the most prominent virus that causes severe respiratory syndrome in human. The nonstructural proteins (nsps) of coronavirus assemble to form the replication transcription complex (RTC) responsible for viral RNA synthesis. In SARS-CoV-2 non-structural protein Nsp8 is a critical component of the RNA replicase and features both well folded secondary structure and disordered segments. It is part of the RNA-dependent RNA polymerase (RdRp) complex and forms a heterotetramer consisting of one molecule of Nsp7, two copies of Nsp8 and one of Nsp12. Nsp8 an Nsp7 are reported to adopt a hollow cylinder-like structure and serve as co-factors for the catalytic Nsp12 that play a role in the stabilization of Nsp12 regions involved in RNA binding and are essential for a highly active Nsp12 polymerase complex.
