1. Bestimmen Sie die Menge an HFN, die zum Beschichten von Kulturgefäßen erforderlich ist, indem Sie die Gesamtoberfläche (cm2) mit der gewünschten Konzentration (μ g/ml) von HFN multiplizieren. Die empfohlene Menge beträgt 2–10 μg/cm2.
2. Die Oberfläche jedes zu beschichtenden Kulturgefäßes mit der Mindestmenge an ausgeglichener steriler Kochsalzlösung (serum- und proteinfrei) befeuchten, die erforderlich ist, um die gesamte Fläche abzudecken.
3. Ggf. die angemessene CO2-Atmosphäre einführen.
4. Jedem Kulturgefäß die berechnete Menge an HFN hinzufügen.
5. HFN 5-20 Minuten lang an der Oberfläche des Gefäßes adsorbieren lassen.
6. Die restliche ausgeglichene Kochsalzlösung vor dem Fortfahren mit Standard-Zellkulturverfahren entfernen.
Das Produkt wird während der Produktion mit einem 0,1-Mikron-Filter filtriert. Es wird empfohlen, das Produkt vor der Verwendung/nach der Rekonstitution erneut zu filtrieren.
