Description de la cible

Complement C5 (UniProt: P01031; also known as C3 and PZP-like alpha-2-macroglobulin domain-containing protein 4) is encoded by the C5 (also known as CPAMD4) gene (Gene ID: 727) in human. The complement system provides innate defense against microbial infection and is a complement to antibody-mediated immunity. It consists of about thirty-five interacting plasma and membrane associated proteins. Activation of the complement system is achieved through three major independent, but overlapping, pathways: the classical pathway, which is activated by certain isotypes of antibodies bound to antigens (immune complexes); the alternative pathway, which is activated on microbial cell surfaces in the absence of antibody; and the lectin pathway, which is activated by a plasma lectin (mannose binding lectin) that binds to mannose residues on microbes. The human C5 complement component is a glycoprotein that is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to produce the mature protein that can further cleave into four chains: Complement C5 beta chain (aa 19-673); Complement C5 alpha chain (aa 678-1676); C5a anaphylatoxin (aa 678-751); and Complement C5 alpha′ chain (aa752-1676). The C5 convertase activates C5 by cleaving the alpha chain, releasing C5a anaphylatoxin and generating C5b. C5a serves as an inflammatory mediator that can activate a variety of cells through G-protein-linked receptors. The C5b fragment combines with C6, C7, C8, and C9 to form lytic C5b-9 complement membrane attack complex (MAC) that is responsible for the membrane lytic events associated with complement activation. Mutations in C5 gene are known to cause complement component 5 deficiency (C5D), a rare defect of the complement classical pathway that leads to susceptibility to severe recurrent infections.
