Description de la cible

Protein Tax-1 (UniProt: P03409; also known as Protein X-LOR, Protein PX, Trans-activating transcriptional regulatory protein of HTLV-1) is encoded by the Tax gene in human. Tax-1 is a homodimeric protein that serves as a transcriptional activator that governs the viral transcription from the 5′LTR via the recruitment of dimers of host phosphorylated CREB1. Together they bind cAMP response elements within the viral promoter and mediate high-level viral transcription. It is reported to shuttle from the host nucleus to the cytoplasm and is found predominantly in the nucleus, where it is equally distributed between the nucleoplasm and the nuclear matrix. Tax-1 is reported to modulate the expression of cellular genes leading to the deregulation of T-cell proliferation, disturbing the integrity of cell cycle checkpoints, the DNA damage response, and apoptotic pathways. It is also shown to act as a ubiquitin E3 ligase and stimulates host IKK complex by catalyzing the assembly of free mixed-linkage polyubiquitin chains, resulting in constitutive activation of NF-kB. Phosphorylation of Tax-1 on threonine 48 is shown to result in the loss of NF-kB activation function. Clone Lt-4 recognizes the Tax-1 protein expressed in HTLV-I-infected cells but does not react with uninfected cell lines and fresh and PHA-activated peripheral blood lymphocytes.
