General description

NF-kappa-B (NF-kB) is a family of ubiquitous transcription factors, which includes RelA, RelB, NF-kB1 and NF-kB2. The NF-kB family regulates multiple cellular processes, including cell growth, differentiation, inflammation, immunity, and apoptosis. In general, inactive forms of NF-kB proteins, bound by IkappaB inhibitory proteins, are present in the cytoplasm, whereas activated forms translocate to the nucleus where they exert transcriptional activity. The p100 subunit is an inactive precursor of NF-kB2, and is proteolyzed to generate the functional p52 protein and enable the formation of an active NF-kB2/p52 dimeric complex. The processing of p100 to form p52 is induced by the NF-kappaB-inducing kinase (NIK), which recruits IkappaB kinase alpha to p100, resulting in phosphorylating of p100 at serines 99, 108, 115, 123, and 872, which is required for subsequent ubiquitination and processing of p100 by the beta-TrCP ubiquitin ligase. Nuclear p100 is phosphorylated at serine 707 by GSK3, which promotes its degradation by Fbxw7α.

Specificity

This antibody recognizes NF-kB (p100) phosphorylated at Ser707.

Immunogen

Epitope: Phosphorylated Ser707
KLH-conjugated linear peptide corresponding to human NF-kB (p100) phosphorylated at Ser707.

Application

Peptide Inhibition Analysis: A 1:500 dilution from a representative lot blocked phospho-NF-kB (p100) (Ser707) in non-starved HEK293 cell lysates.
