Specificità

Clone 2M5 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects CYP11A1. It targets an epitope within 24 amino acids from the N-terminal half.

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal half of human CYP11A1.

Applicazioni

Quality Control Testing

Evaluated by Western Blotting in human placenta tissue lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CYP11A1 protein in human placenta tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000-10,000 dilution from a representative lot detected CYP11A1 in human adrenal and human testis tissue lysate.

Affinity Binding Assay: A representative lot of this antibody bound CYP11A1 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected CYP11A1 in human adrenal gland tissue sections.

Immunohistochemistry (Paraffin) Analysis (IHC(P)): A 1:1,000 dilution from a representative lot detected CYP11A1 in human adrenal gland tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Cholesterol side-chain cleavage enzyme, mitochondrial (UniProt: P05108; also known as EC:1.14.15.6, CYPXIA1, Cholesterol desmolase, Cytochrome P450 11A1, Cytochrome P450(scc)) is encoded by the CYP11A1 (also known as CYP11A) gene (Gene ID: 1583) in human. This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases that catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the mitochondrial inner membrane and catalyzes the conversion of cholesterol to pregnenolone, the first and rate-limiting step in the synthesis of the steroid hormones. Two transcript variants encoding different isoforms have been found for this gene. The cellular location of the smaller isoform is unclear since it lacks the mitochondrial-targeting transit peptide. Its highly expressed in steroidogenic tissues such as the adrenal cortex, ovaries, and testes. Its expression is regulated by trophic hormones like ACTH in the adrenal cortex and LH in the gonads. Mutations in the CYP11A1 gene can lead to defects in steroid hormone biosynthesis, resulting in CAH, a condition characterized by impaired adrenal function. Altered expression of CYP11A1 has been implicated in the pathogenesis of PCOS, affecting steroid hormone levels. Studies show that artemisinins promoted cytochrome P450 family 11 subfamily A member 1 (CYP11A1) protein degradation to block androgen overproduction. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Stato fisico

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Ricostituzione

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stoccaggio e stabilità

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
