General description

Focal adhesion kinase (FAK) or PTK2 (protein tyrosine kinase 2) is a tyrosine kinase, which resides in the cytoplasm, and concentrated at cellular focal points. It is a highly conserved 125kDa protein, which has a wide range of tissue expression. It is composed of 1,052 amino acids, and contains N- and C-termini flanking the central kinase domain. The N-terminal called the FERM (four-point-one, ezrin, radixin, moesin) domain, controls FAK activity, whereas the C-terminal contains the focal-adhesion-targeting (FAT) domain.[1]

Biochem/physiol Actions

Focal adhesion kinase (FAK) or PTK2 protein tyrosine kinase 2 plays multiple roles such as, control of transcription, cell cycle progression, apoptosis, cell migration, and metastasis in case of tumor cells. This attributes to its predominant role in cell signaling. It is involved in the infection of rabies virus (RABV).[1] Studies in Spanish population show that polymorphisms rs7460 and rs7843014 in this gene might be linked with exceptional longevity.[2] Phosphatidylinositol 4-phosphate 5-kinase type Iγ enzyme produces phosphatidylinositol-4,5-bisphosphate [PI(4,5)P2] in focal adhesion, which cross-links FAK activation to integrin signaling.[3]

Supplied in 50mM NaPhosphate pH 7.0, 300 mM NaCl, 150 mM imidazole, 0.1mM PMSF, 0.2 mM DTT, 25% glycerol.
