Specificity
Clone 1J2 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Growth factor receptor-bound protein 2 (GRB2). It targets an epitope within 15 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal half of human Growth factor receptor-bound protein 2 (GRB2).
Application
Quality Control Testing
Evaluated by Western Blotting in HEK293 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected GRB2 in HEK293 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GRB2 in HeLa, PC12, and RAW264.7 cell lysates.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GRB2 in HEK293 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected GRB2 in Human tonsil tissue sections.

Target description
Growth factor receptor-bound protein 2 (UniProt: P62993; also known as Adapter protein GRB2, Protein Ash, SH2/SH3 adapter GRB2) is encoded by the GRB2 (also known as ASH) gene (Gene ID: 2885) in human. GRB2 is a signaling adaptor that is ubiquitously expressed and interacts with numerous receptors and intracellular signaling molecules. It contains one central SH2 domain flanked by two SH3 domains and is broadly expressed in many tissues and is shown to be essential for embryonic development and multiple cellular functions. It is a major mediator in Ras-MAPK activation pathway induced by numerous receptors because of its association with son of sevenless (SoS). GRB2 plays a role in early T cell development and is shown to be a positive regulator of TCR signaling. It does not directly affect the activities of Ras and Erk1/2; however, it amplifies the activity of Lck, a tyrosine kinase that functions at the top of the TCR signaling cascade. It exists in a constitutive equilibrium between monomeric and dimeric states. The monomeric form is capable of binding to SoS and upregulating MAP kinase signaling, and the dimeric state is inhibitory to this process. Phosphorylation of tyrosine 160 or binding of a tyrosylphosphate-containing ligand to the SH2 domain of Grb2, results in dimer dissociation. Phosphorylation of tyrosine 160 is readily detectable in the malignant prostate, colon. and breast cancers. Two isoforms of GRB2 have been described that are produced by alternative splicing. Isoform 2 acts as a dominant negative protein that suppresses proliferative signals, may trigger active programmed cell death. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
