Spécificité

Clone 3F13 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects TANK-binding kinase 1-binding protein 1 (TBKBP1). It targets an epitope within 17 amino acids from the C-terminal region.

Immunogène

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human TANK-binding kinase 1-binding protein 1 (TBKBP1).

Application

Quality Control Testing

Evaluated by Western Blotting in NTERA-2 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TBKBP1 in NTERA-2 cell lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound TBKBP1 peptide with a KD of 2.1 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TBKBP1 in human placenta tissue sections.

Western Blot Analysis: A 1:1000 dilution of a representative lot of the this antibody detected TBKBP1 in HEK293T cells transfected with a TBKBP1 expression vector.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Description de la cible

TANK-binding kinase 1-binding protein 1 (UniProt: A7MCY6; also known as TBK1-binding protein 1) is encoded by the TBKBP1 (also known as KIAA0775, SINTBAD) gene (Gene ID: 9755) in human. TBKBP1 is an adapter protein that constitutively binds TANK-binding kinase 1 (TBK1) and Inhibitor of NF-kB kinase subunit epsilon (IKBKE) and plays a role in antiviral innate immunity. Its TBK1 and IKBKE interacting region is localized in amino acids 280-329. It can exist as a homodimer or can form heterodimer with NAP1. Its homodimerization region is localized in amino acids 1-279. TBKBP1 expression is observed in leukocytes, lung, placenta, small intestine, liver, kidney, spleen, muscle, heart, and brain. TBK1 that is reported to be a central mediator of growth factor signaling relies on TBKBP1 for its action. TBKBP1 recruits TBK1 to protein kinase C (PKC ) through a scaffold protein, caspase recruitment domain 10 (CARD10). This enables PKC to phosphorylate TBK1 at serine 716. This phosphorylation is essential for TBK1 activation by growth factors, but not by innate immune stimuli. It has been reported that although the TBK1-TBKBP1 signaling axis is not essential for the induction of type I interferon, it facilitates mTORC1 activation and oncogenesis. Deletion of either TBK1 or TBKBP1 is known to inhibit tumorigenesis in a murine model of lung cancer. The TBK1-TBKBP1 axis can also facilitate tumor-mediated immunosuppression via induction of PD-L1 and stimulation of glycolysis. Two isoforms of TBKBP1 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
