Description générale

Fas-associated protein 1 (FAF1) is a member of the TNF receptor superfamily that plays a critical role in apoptosis during development and immune function. FAF1 has two amino-terminal domains with structural homology to ubiquitin. FAF1 has been observed to be one of the components of the death-inducing signaling complex (DISC) during Fas-mediated apoptosis and can bind to caspase-8 and FADD. Additionally, FAF1 has been shown to suppress the activation of the NF-kappaB transcription factor. FAF1 is also known to interact with the inflammatory signaling PYRIN-containing Apaf-1-like proteins (PYPAFs, also called NALPs) such as PYPAF1, PYPAF2 (NALP2), and PYPAF7, suggesting FAF1 may also be involved in the inflammation pathway. FAF1 is widely expressed with highest expression observed in testis, slightly less abundant in skeletal muscle and heart, followed by prostate, thymus, ovary, small intestine, and colon.
