Specificity

Clone 2K15 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects Transcription factor SOX-4 (SOX4). It targets an epitope within 16 amino acids from the N-terminal half of the protein.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the N-terminal half of human Transcription factor SOX-4.

Application

Quality Control Testing

Evaluated by Western Blotting in human brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Transcription factor SOX-4 in human brain tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Sox4 in SOX-4 Recombinant Protein.

Affinity Binding Assay Analysis: The binding affinity of this antibody to Sox4 peptide was at least ten thousand-fold (10,000X) higher than that obtained with the non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Sox4 in A-431, HeLa, and NIH/3T3 cells.

Target description

Transcription factor SOX-4 (Uniprot: Q06945; also known as SOX4) is encoded by the SOX4 gene (Gene ID: 6659) in human. This intronless gene encodes a member of the SOX (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. SOX4 is mapped to human chromosome 6p22. It also acts as a transcriptional activator that binds with high affinity to the T-cell enhancer motif 5′-AACAAAG-3′ motif. Acetylation of SOX4 at Lys-95 by KAT5 is reported to promote the transcription activator activity which is required during myoblast differentiation. This acetylation abolishes the interaction between SOX4 and HDAC1 and switches SOX4 into a transcriptional activator. SOX4 is frequently over-expressed in several malignancies due to gene amplification and activation of PI-3 kinase, Wnt, and TGFβ signaling pathways. Knockdown of SOX4 gene is reported to reduce migration of cancer cells in various murine models. SOX4 is primarily associated with embryonic development, where it participates in the formation of various tissues and organs. It also plays a major role in modulating RELN (reelin) expression and the inside-out pattern of cortical layer formation. It is essential for the establishment of pan-neuronal protein expression. The protein may also function in the apoptosis pathway leading to cell death as well as to tumorigenesis and may mediate downstream effects of parathyroid hormone (PTH) and PTH-related protein (PTHrP) in bone development. The solution structure has been resolved for the HMG-box of a similar mouse protein. Dysregulation of SOX4 has been implicated in several diseases, including cancer, where it can act as an oncogene, tumor suppressor or can contribute to tumor progression. Furthermore, research on SOX4 is essential to better understand its molecular mechanisms and its potential as a therapeutic target. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
