General description
Ras proto-oncogene is a guanine-nucleotide binding protein that couples tyrosine kinase receptor signals to mitogen activated protein (MAP) kinase cascade. It has an important role in cell growth. The Ras gene is localized on human chromosome 11p15.5. Ras is a substrate for farnesyltransferase (FTase) and when farnesylated, it is anchored to the membrane.
Application
Human Ras has been used as a negative control in tubulin overlay assay.
Biochem/physiol Actions
Ras proteins, p21, are guanyl nucleotide binding proteins that play an important role in cellular signal transduction. Ras proteins are regulatory proteins that bind GTP in their active state. A Ras intrinsic GTPase activity hydrolyzes guanosine-5′-triphosphate (GTP) to guanosine diphosphate (GDP) thus inactivating Ras, while activation of Ras requires exchange of the bound GDP with GTP. Inhibition of Ras activity by stimulation of its GTPase activity is enhanced by GTPase activating proteins. Membrane localization of Ras is crucial for its cellular activity. Inhibition of Ras farnesylation is a target for anti-cancer drug development, since abnormally active Ras is present in more than 50% of human cancers.
