Gene Information

Specificity

Serotonin Transporter (SERT). An antibody raised against a similar peptide has been shown to react with a SERT in baculovirus cells expressing recombinant rat and human SERT. Antibody activity may be reduced considerably when detecting endogenous SERT from rat or human cells. SERT has been observed to migrate as 94 kDa (rat platelet), 80 kDa (pulmonary membranes) and 76 kDa (rat brain). This could be due to differences in postranslational membrane modifications (e.g. glycosylation).

Immunogen

A 15 amino acid sequence between transmembrane domain 7 and 8 of the rat serotonin transporter. This region is presumably from the extracellular domain.

Cell pellets are resuspended in 100 μL lysis buffer (25 mM Tris, pH 7.6, 150 mM NaCl, 1 μg/mL aprotonin, 10 μg/mL leupeptin, 1 mM phenylmethylsulfonylfluoride, 1 mM sodium orthovanadate, 1 mM EDTA, and 1% Triton X-100). Samples were incubated for 1 hour at 4°C and were pelleted at high speed for 10 minutes at 4°C, and 100 μg protein per sample was subjected to 10% sodium dodecyl sulfate-polyacrylamide gel electrophoresis.





ELISA: 1:100,000 primary EIA using immunogen only (50-100 ng SERT peptide (Catalog Number AG297)/well). Not tested on purified protein at the stated dilutions.


Optimal working dilutions must be determined by end user.

Physical form

Affinity purified immunoglobulin. Liquid in 100 mM Tris, pH 7.5 with 0.2% BSA and 0.05% sodium azide.

Storage and Stability

Maintain at -20°C in undiluted aliquots for up to 12 months. Avoid repeated freeze/thaw cycles.
